肌动蛋白和肌球蛋白结合蛋白参与调节平滑肌和非肌细胞中的肌动球蛋白相互作用（可能充当肌球蛋白和肌动蛋白丝之间的桥梁）。刺激原肌球蛋白的肌动蛋白结合，从而增加肌动蛋白丝结构的稳定性。在肌肉组织中，通过与F-肌动蛋白结合来抑制放线菌素ATPase。该抑制被钙钙调蛋白减弱，并被原肌球蛋白增强。与肌动蛋白，肌球蛋白，两个原肌球蛋白分子和钙调蛋白相互作用。在细胞有丝分裂和受体加帽过程中也起着至关重要的作用。
