STING分子在最初发现时只被认为是CDS信号通路中的一个普通接头蛋白,进一步研究表明它是环二核苷酸cyclic dinucleotide(CDNs)的直接感受分子。CDNs是细菌合成的重要第二信使,调节原核生物及哺乳动物的众多生物进程,能启动宿主天然免疫反应。CDNs和山酮衍生物xanthenone derivative(例如DMXAA),可结合并活化STING,通过TBK1-IRF3依赖的信号通路来诱导I型干扰素表达。
