TLR7和TLR8都是与病毒感染引起的免疫反应相关的胞内受体。它们识别富含GU的短单链RNA,以及咪唑喹啉和核苷类似物等人工合成的小分子。TLR7/TLR8的信号通路依赖于MyD-88的活化、AP-1和IFN-α/β等转录因子入核,以及IRF7的磷酸化。IRF7磷酸化诱导ISRE的表达和I型干扰素基因的产生,而AP-1和IFN-α/β的激活导致细胞因子的释放。

细胞表面受体TLR5可识别鞭毛蛋白,后者是革兰氏阳性菌和革兰氏阴性菌表面鞭毛丝的主要成份。TLR5活化后通过MyD88信号通路促进一些炎症因子的释放。TLR5通过形成同源二聚体来激活炎症信号通路,这显示它可能是唯一参与鞭毛蛋白识别的TLR分子。

TLR13是在小鼠中发现的一个胞内受体。目前关于它的配体和信号通路仍知之甚少,但23S核糖体RNA(rRNA)已被证明是它的配体之一。TLR13诱导的细胞因子释是透过依赖MyD-88和UNC93B这两个分子来激活AP-1和INF-α/β等转录因子入核。TLR13也可通过活化IRF7来诱导IFN-γ的产生。人类中不存在TLR13受体,可能是通过其它类似的病原受体来发现病原菌的入侵。

NOD1(又称CARD4)和NOD2(又称CARD15)是能感知细菌肽聚糖(PGN)成份的细胞内模式识别受体。NOD1识别全部革兰氏阴性菌和部分革兰氏阳性菌PGN成份中的iE-DAP,而NOD2则识别差不多全部细菌都带有的MDP结构。iE-DAP和MDP成份必须通过细菌侵入细胞或其它细胞物质摄入方式进入到细胞后才能被识别。识别配体后,NOD1和NOD2形成多聚体,通过RIP2激酶的丝氨酸/酪氨酸来传递讯号。激活后的RIP2可泛素化修饰NEMO/IKK©,从而引起NF-kB的活化和炎症因子的释放。

Dectin-1是C型凝集素受体(CLR)家族成员之一,在抗真菌天然免疫反应中发挥重要作用。Dectin-1是β-glucan(β-葡聚糖)的特异性跨膜受体。β-glucan,即葡萄糖聚合物,则是真菌(包括酿酒酵母Saccharomyces cerevisiae和白色念珠球菌Candida albican)细胞壁的主要成分。与其受体结合后,Dectin-1会引起细胞胞吞作用,并通过Syk激酶和接头分子CARD9/Bcl10/Malt1传递信号,进而诱导活性氧ROS的产生、NF-κB的活化及后续炎症因子的产生。Dectin-1和TLR2可协同发挥作用来增强各自的信号级联反应。

Mincle是一个多功能的危险信号感知分子,可识别一系列配体,包括受损的细胞、真菌、酵母和结合分枝杆菌。Mincle的胞外配体包括真菌的α-mannose(甘露糖)、分枝杆菌的糖脂和来自结核分枝杆菌具有免疫刺激功能的TDM(trehalose-6’6’-dimycolate)组分。此外,Mincle还可通过识别其内源的DAMP来感知受损的细胞。SAP130(剪接体相关蛋白130)是已被发现的DAMP之一,它是一种由坏死细胞释放的可溶性因子。Mincle通过Syk激酶传递信号,透过CARD9来激活NF-κB。Syk还可引起胞内的钙动员和calcineurin-NFAT信号通路的活化。
