Thermo Scientific EZ-Link Sulfo-NHS-SS-Biotin enables simple and efficient biotin labeling of antibodies, proteins and any other primary amine-containing biomolecules in solution.

Sulfo-NHS-SS-Biotin is a thiol-cleavable amine-reactive biotinylation reagent that contains an extended spacer arm to reduce steric hindrances associated with avidin binding. This reagent is water-soluble, enabling biotinylation to be performed in the absence of organic solvents such as DMSO or DMF for applications that cannot tolerate solvents or are complicated by their inclusion.

Highlights:

  • Label almost any protein with biotin to facilitate immobilization, purification or detection
  • Ideal for reversible biotinylation; disulfide bond in the spacer arm allows the biotin label to be removed as desired
  • Amine reactive Sulfo-NHS ester reacts rapidly with any primary amine-containing molecule to attach the biotin label via a stable amide bond
  • Sulfonate group attached to the biotinylation reagent increases water solubility to a minimum of 6 mg/ml, so biotin labeling can be done under physiologic conditions
  • Long spacer arm on the biotin reduces steric hindrance when binding to avidin molecules

Thermo Scientific Pierce Streptavidin and Conjugates include purified recombinant streptavidin biotin-binding protein, plus standard SA-HRP, SA-AP and fluorescent conjugates for biotin-based assays.

 

These purified and conjugated forms of recombinant streptavidin protein enable detection of biotinylated antibodies and other probes in a variety of standard assay methods, including Western blotting, ELISA, immunohistochemistry and fluorescence imaging. Purified streptavidin and the streptavidin-HRP conjugates （peroxidase-conjugated streptavidin） are supplied lyophilized in phosphate-based buffers for immediate reconstitution with water. Alkaline Phosphatase conjugates are supplied as a liquid in a Tris-based buffer.

Streptavidin is a tetrameric protein containing four biotin-binding sites similar to avidin. The native protein isolated from culture filtrates of Streptomyces avidinii is carbohydrate-free, has an acidic pI of 5.5 and a mass of 75 kDa. Streptavidin generally has less nonspecific binding than avidin （from chicken egg white） because of the absence of carbohydrates.

Our Streptavidin Resins （i.e., Immobilized Streptavidin Products） use a recombinant form of streptavidin with a mass of 53 kDa and a near-neutral pI. These resins are prepared by covalently coupling streptavidin to the porous agarose or UltraLink Biosupport using efficient and stable chemistries, resulting in Streptavidin Agarose and UltraLink Resins that are leach-resistant and stable at pH 2-11. The products are excellent choices for a variety of small- or large-scale affinity purification applications involving biotinylated macromolecules, including separation of biotinylated molecules from samples and immunoprecipitation of antigens using biotin-labeled antibodies.

   Thermo Scientific Detoxi-Gel Endotoxin Removing Gel and Columns provide a quick method for removal of endotoxins （lipopolysaccharides or LPS） from samples. Even low levels of endotoxins can be toxic to cells or organisms and must be removed before biological samples can be introduced.

Thermo Scientific Pierce TPCK Trypsin is a serine endoprotease that is applicable to amino acid analysis and protein sequencing, mapping and structural studies; the immobilized form allows sample separation after treatment.

Trypsin has a wide range of applications including amino acid analysis and protein sequencing, mapping and structural studies. Enzymes such as trypsin and chymotrypsin have become important tools in sequencing studies since they are highly selective in their cleavage of peptide bonds. Trypsin cleaves only those peptide bonds in which the carboxyl group is contributed by a lysine or an arginine residue, regardless of the length or amino acid sequence of the chain.

Immobilized trypsin can be substituted for free trypsin in any application, and is advantageous because it virtually eliminates autolysis, eliminates contamination of a sample with the protease and allows control of the digestion by removing the trypsin. Immobilized trypsin is also more stable against heat-induced denaturation, resulting in longer maintenance of activity.
