The Thermo Scientific Pierce Activated Peroxidase and Antibody Labeling Kit is designed to prepare and purify HRP-antibody conjugates with any previously unmodified primary or secondary antibody. Activated Peroxidase is HRP enzyme that has been modified with glutaraldehyde to convert surface amines to active aldehydes groups that will spontaneously conjugate with amines on an antibody. The result is a conjugate where both the enzyme activity and the antigen binding activity are preserved. The Pierce Activated Peroxidase Kit includes a Protein A/G column and buffers to affinity-purify the antibody conjugate from excess, unreacted enzyme. Highlights: Reacts with primary amines to form covalent amide bonds; no reduction step is necessary to secure the linkage Activated enzyme can be stored for at least 12 months at -20°C One milligram of activated enzyme is sufficient to produce about 0.5mL of conjugate with a working dilution of 1:1000 when coupled to a high-titer antibody Enzyme activity is greater than 200 units/mg

Thermo Scientific Pierce Plus Activated Peroxidase is an amine-reactive form of horseradish peroxidase (HRP) that provides coupling efficiencies of greater than 95% with antibodies and other proteins! Sugar groups on native HRP have been oxidized with periodate to produce amine-reactive aldehydes. The activated peroxidase spontaneously and efficiently crosslink with primary amines on antibody or other proteins. The method is more effective than other amine-reactive chemistries, such as glutaraldehyde coupling, which often causes polymerization and conjugate inactivation. This high-quality prepared enzyme eliminates the difficulties inherent in preparing and validating activated peroxidase from scratch. The kit provides the enzyme, accessory reagents and protocol to easily produce high-performance conjugates for Western blotting, ELISA and other detection techniques.
