Thermo Scientific SuperSignal West Pico Chemiluminescent Substrate is an enhanced chemiluminescent (ECL) substrate for horseradish peroxidase (HRP) enzyme that provides low picogram detection of proteins in Western blot applications.

SuperSignal West Pico Substrate emits light for longer and chemiluminescent signal at more than twice the intensity of other entry-level luminol-based horseradish peroxidase detection systems while also maintaining sensitivity and low background. In addition, the greater signal intensity is achieved using less primary and secondary antibody than typical ECL substrates. All of this for about 30% less cost than comparable products.

Highlights:

  • ECL substrate – an enhanced chemiluminescent substrate for detection of horseradish peroxidase (HRP) activity from antibodies and other Western blot probes
  • Economy – costs less per milliliter than other similarly sensitive chemiluminescent substrates
  • Picogram sensitivity – highly sensitive for the rapid development of a wide range of protein levels
  • Stable light output – long-duration signal makes it possible to make multiple exposures to film

The Thermo Scientific Pierce Secondary Antibodies featured here are affinity-purified polyclonal antibodies with well-characterized specificity for mouse immunoglobulin classes (IgG, IgM, IgA).

Each of these anti-mouse immunoglobulin polyclonal antibodies is affinity-purified using its target antigen (i.e., the target mouse primary antibody class, subclass and fragment) coupled to an agarose support. Affinity purification eliminates nonspecific antibodies, resulting in high specificity and low background. The purification process involves an elution procedure, that yields antibodies with high avidity. Conjugated antibodies are affinity-purified before being labeled and contain bovine serum albumin as a stabilizer.

This purification is accomplished by adsorbing the antigen-purified antibody to an affinity column containing immobilized serum proteins of selected species. This additional processing minimizes antibody cross-reactivity to the serum of proteins of the species indicated by the code “min x Sp Sr Prot,” where the cross-absobed species are indicated by the accompanying abbreviation key.

 IFN-γ is an acid-labile interferon produced by CD4 and CD8 T lymphocytes as well as activated NK cells. IFN-γ receptors are present in most immune cells, which respond to IFN-γ signaling by increasing the surface expression of class I MHC proteins. This promotes the presentation of antigen to T-helper （CD4+） cells. IFN-γ signaling in antigen-presenting cells and antigen-recognizing B and T lymphocytes regulate the antigen-specific phases of the immune response. Additionally, IFN-γ stimulates a number of lymphoid cell functions including the anti-microbial and anti-tumor responses of macrophages, NK cells, and neutrophils. Human IFN-γ species-specific and is biologically active only in human and primate cells. Recombinant human IFN-γ is a 16.7 kDa protein containing 143 amino acid residues.
