TRF分子量约7.7万,为单链糖蛋白,含糖量约6%。TRF可逆地结合多价离子,包括铁、铜、锌、钴等。每一分子TRF可结合两个三价铁原子。TRF主要由肝细胞合成,半衰期为7天。血浆中TRF的浓度受铁供应的调节,在缺铁状态时,血浆TRF浓度上升,经铁有效治疗后恢复到正常水平。
血浆中TRF水平可用于贫血的诊断和对治疗的监测。在缺铁性的低血色素贫血中TRF的水平增高(由于其合成增加),但其铁的饱和度很低(正常值在30%-38%)。相反,如果贫血是由于红细胞对铁的利用障碍(如再生障碍性贫血),则血浆中TRF正常或低下,但铁的饱和度增高。在铁负荷过量时,TRF水平正常,但饱和度可超过50%,甚至达90%。
TRF在急性时相反应中往往降低。因此在炎症、恶性病变时常随着白蛋白、前白蛋白同时下降。在慢性肝疾病及营养不良时亦下降,因此可以作为营养状态的一项指标。
有免疫试剂盒供应抗体级标准品。用免疫扩散或浊度法检测。正常成人参考值为2200-4000mg/L。新生儿为1300-2750mg/L。临床评价时常同时测定血清铁含量及TRF的铁结合容量(TIBC),并可计算出的TRF铁饱和度(%)。TRF亦可通过测定而间接计算估得,其计算方程式如下:

动物实验 | 动物室环境和光周期的条件是20-25℃,40%-70%湿度和12小时光照/ 12小时黑暗循环。每只小鼠在右侧皮下接种MV-4-11肿瘤细胞(5×106)。当肿瘤大小达到80-150 mm3时,治疗开始。根据肿瘤大小将小鼠随机分为治疗组;使用卡尺在2维中测量肿瘤大小,并且使用以下公式以mm 3表示体积:V = 0.5 a×b2其中a和b分别是肿瘤的长径和短径。预处理随机化确保每组具有大致相同的平均肿瘤大小。用载体(5%右旋糖),SGI-1776或阿糖胞苷(ara-C)处理小鼠。 SGI-1776和ara-C以5%葡萄糖配制。 SGI-1776通过口服强饲法(PO)以每日×5 /周或每周两次的方案施用; ara-C通过腹膜内注射3次/周给药,连续3周。当测量的肿瘤大小大于3000mm 3或当它们失去≥20%的初始体重时,对动物实施安乐死;如果体重减轻≥15%,则首先停止治疗直至小鼠恢复体重。即使停止治疗,当体重减轻仍然≥20%时,对小鼠实施安乐死。 T / C值(以%表示)是抗肿瘤效力的指示,其中T和C分别是给定日的治疗组和对照组的平均肿瘤体积。 |

用法
称取本品 31.0g,加热溶解于 1000ml 蒸馏水,分装试管,每管 3ml,121℃高压灭菌 15 分钟备用。
注:培养基中含少量硫酸亚铁,灭菌后可能出现微量沉淀。
硫化氢试验原理
有些细菌可分解培养基中含硫氨基酸或含硫化合物,而产生硫化氢气体,硫化氢遇铅盐或低铁盐可生成黑色沉淀物。
