■ 制品说明
本制品是利用PCR技术进行定点突变操作的试剂盒。其原理是利用导入变异点的引物进行PCR反应后，对PCR产物进行末端平滑化及5’磷酸 (P) 化处理，再用高效连接试剂Ligation Solution I进行自身连接 (环化反应)，然后转化、提取突变体DNA。
本试剂盒的原理十分简单，但Taq酶在PCR过程中的错配率限制了这一技术的应用。现在，Takara的Pyrobest DNA Polymerase使这一技术得到了很好的发挥。Pyrobest DNA Polymerase不仅具有很高的保真性（Fidelity），并且和TaKaRa Taq具有同样的扩增效率。与其他突变试剂盒相比，本试剂盒的特点是操作方便、简单，只需一天时间便可完成整个操作，并且不受载体、宿主菌的限制。

■ TaKaRa MutanBEST Kit原理
试剂盒原理见下图，全套操作约需5小时，简单说明如下。
1. 设计一对5’端邻接，3’端方向相反的引物用以导入变异点。
2. 进行PCR扩增 (使用高保真DNA聚合酶Pyrobest DNA Polymerase)。
3. PCR片段的末端平滑及5’末端磷酸化反应 (在同一反应体系内进行，反应只需10分钟)。
4. 自身连接反应 (使用本试剂盒中的DNA高效连接液Ligation Solution I，连接反应只需30 分钟至1小时)。
5. 质粒DNA转化，挑选变异体。

■ 注意事项
1. 当载体与插入的DNA片段的总长度小于5 kb时，使用本试剂盒较为合适；如果载体与插入DNA 片段长度太长时，不推荐使用本试剂盒。
2. PCR反应后的DNA片段应进行切胶回收纯化，然后再进行自身连接反应，这样可以提高突变成功率。
3. Ligation Solution I请于冰中融解，使用前混匀。应避免多次反复冻融。
4. 连接转化效率低时，请试用下述方法进行改进。
① 确认感受态细胞的转化效率。
② 延长连接反应时间至过夜反应。

胶原酶（Collagenase）是一种蛋白酶,能够特异性的识别 Pro-X-Gly-Pro 序列并切割该
序列中性氨基酸（X）和甘氨酸（Gly）之间的肽键。许多蛋白酶都能水解单链且变性的胶原多肽,但胶原酶是唯一一种可以降解具有三股超螺旋结构的天然胶原纤维的蛋白酶,这种胶原纤维广泛存在结缔组织内。胶原酶一般分为 I、II、III、IV 型以及肝细胞专用胶原酶,要根据所要分离消化的组织类型选择胶原酶类型。
1、胶原酶 I 用于上皮、肺,脂肪和肾上腺组织细胞的分离。
2、胶原酶 II 适用于脂肪,肝脏,骨,甲状腺,心脏和唾液腺组织。
3、胶原酶 III 包含至少 7 种蛋白酶成分,分子量从 68-130KD 不等。它能消化多种组织。
4、胶原酶 IV 包含至少 7 种蛋白酶成分,分子量从 68-130KD 不等。它可用于胰腺小岛组织的分离,将结缔组织分离成单个细胞。

使用方法：（根据实际需要参阅相关文献配制和使用）
取胶原酶中加入到相应溶解液(根据特定的实验条件或者参考相应的文献资料确定所需的最佳工作浓度和体积),轻轻旋涡震荡使其充分溶解,然后用低蛋白结合性的 0.22um 的滤膜过滤除菌即可使用,或于分装后-20℃避光保存,避免反复冻融。酶液在含有 0.3-0.5mM 钙离子 PH7.4 的条件下可以在 37℃维持活性 5 小时。在-20℃可维持数月。
