检验原理
蛋白胨提供碳源和氮源；葡萄糖提供能源；磷酸二氢钾为缓冲剂；硫酸镁提供必须的微量元素；玫瑰红钠作为选择性抑菌剂可抑制细菌的生长，并可减缓某些霉菌因生长过快而导致菌落漫延生长；琼脂是培养基的凝固剂。

按标签用法制备培养基，接种以下质控菌株，放置20-25℃需氧培养48-72小时。

分装及储存：
1、在进口原装抗体分装时,加入50%灭菌的甘油,能延长试剂的保存期。但应注意的是,此时抗体的浓度及稀释比例应降低一倍。
2、初次使用参考推荐的稀释比例,实际实验操作过程中需根据抗原和抗体的具体情况适当调节二抗的稀释比例。但应注意,需现用现配,因为稀释后抗体的有效活性会很快降低。
3、-20℃保存,避免反复冻融。
