为确保内切酶发挥 100% 的酶切活性,NEB公司随酶免费提供 10X 反应缓冲液(NEBuffer)。绝大多数内切酶配有四种标准反应缓冲液中的一种,但有些内切酶配有特殊的反应缓冲液。四种标准反应缓冲液具有不同颜色标记,以便实验者使用方便。

产品简介:

本产品专门用于抑制和清除水浴锅中的微生物,多种高效清除成分能够有效抑制清除影响细胞培养的各类细菌、真菌及其芽孢和孢子、支原体、病毒等微生物。本产品无色无味,作用完毕后分解为水和氧气,没有任何毒性和毒副产物,可替代传统的硫酸铜等易腐蚀的消毒试剂,可适用于水浴锅的日常杀菌处理。

 

使用方法:

1、首先将水浴锅内的水盘清理干净,建议使用细胞房除菌剂(BL1078A)或者75%酒精等消毒溶液擦拭表面。

2、在处理过的水浴锅中加入正常体积的水,按照1:1000比例加入水盘除菌剂(例如10L水加入10ml水浴锅除菌剂)

3、需要再添加水时,按照同样的比例加入水浴锅除菌剂即可,

4、建议7-10天更换一次。

 

注意事项

1、 本产品安全无毒,无刺激性,但是在使用过程中建议佩戴口罩,避免和皮肤黏膜接触,如果接触请立即用大量清水冲洗

DEAE-纤维素,它采用平均粒径为50µm的颗粒型亲水高分子聚合物,表面又用大分子糖链接枝,使它有更高的比表面积和更好的生物兼容性,保持更高载量,同时又具有更好的分辨率。由于比表面积大,平衡和洗脱的时间也更短。它经过接枝即使是纯化病毒,质粒等超大分子的物质,载量基本保持不变。

注意事项:

1、 色谱柱装填

(1)所需要用到的材料的温度要与色谱操作的温度一样,液体最好做脱气处理。填料可直接称量需要的量用缓冲液溶涨一小时装柱即可,如果不好溶胀,可适当加热。

(2)在柱子下端加入20%乙醇,以除去柱子中的空气,关闭柱子出口,在柱内保留少量的20%乙醇。20%乙醇容易产生气泡,可以在里面加1%吐温避免气泡产生。也可以换成纯水装柱子,但是需要把填料中的20%乙醇也换成纯水,具体的方法取需要体积的填料在抽滤漏斗上进行,也可以小心倾去填料上的20%乙醇,再换成5倍体积的纯水,反复沉淀去上清,5次左右就可以用于装柱子。

(3)此填料颗粒比较细,所以一定要注意柱子要选择合适的筛网,不能漏,也可以取点填料加到筛网上试试,如果没有问题再将填料连续倒入柱子时,要用玻璃棒的紧靠柱子内壁引流,以减少气泡的产生,让填料先自然沉降到填料体积不再变化,而填料和上面的液体很好分层,上层溶液完全澄清,就可以开泵用适当的流速压柱子,填料体积不再变化后,再把转换头紧顶在填料上就可以平衡柱子使用。使用的流速要小于装柱子的流速。

(4)在装柱子前,填料从冰箱中取出至少要室温放置2-3个小时,这样避免装柱子时由于温度变化而使柱子中产生气泡。

2、 蛋白的结合

样品的盐浓度和pH要尽量和平衡柱子的缓冲液一致,盐浓度过高或者pH带低也许挂不上,所以要根据自己的样品做适当调整。

3、 蛋白的洗脱

这个填料如果采用线性梯度洗脱,柱子的直径和高度比最好是大于10,数值越大越有利于分离,而且样品最好别上太多,可以按约10mg/ml上样,如果采用阶段洗脱的方法,装短粗柱子就可以,上样量也没有限制。阶段洗脱容易放大,重复性好,如果洗脱条件好完全可以得到和线性梯度一样或者更好。采用什么方法完全根据自己需要。

4、 再生清洗

(1)用完之后用0.1M 醋酸洗5个柱体积,然后用2M NaCl洗5个床体积,再用水洗至中性,然后用20%乙醇保存。

(2)有机溶剂和水混合很容易产生气泡,为了避免这样情况,可以把配好的有机溶剂在室温放置过夜,再使用,这样可以避免气泡进柱子而导致柱子不能正常使用。

特别注意:
上样之前,样品至少用0.45微米膜过滤,尽量去除色素,否则色素会被吸附到填料上,影响填料的正常使用。
在使用过程中,不能使用强酸强碱,酸和碱的浓度应低于0.15摩尔。

注意事项

1: 上述培养基含有各种所需成分(葡萄糖除外,因为有些研究者需要用其他糖类如用半乳糖诱导GAL1启动子表达),为各种氨基酸和酵母含氮碱/酵母氮源的精细混合物干粉,按照8g/L称取,加水后即可配制成相应的培养液,无需再购置添加Minimal SD Base或其他酵母氮源成分。高压灭菌后加无菌葡萄糖至终浓度为2%(或棉子糖)或实验所需的特定诱导物(如半乳糖用于GAL1启动子诱导表达的剂量或浓度为2%)即可,适用于外源基因在酵母中的表达,酵母单杂交(yeast one-Hybrid)和酵母双杂交(yeast two-Hybrid)系统等的选择缺陷型培养,筛选含特定标志基因的酵母表型。

2: 在配制酵母缺陷培养基时,可以先加入葡萄糖后再高压,对酵母的生长不会产生严重影响。但如加入葡萄糖后再高压消毒则培养基会变色,由浅黄到深褐色不等,对细胞的生长还是有一定影响的(但在一般情况下不会影响实验)。建议: 由于氨基酸和葡萄糖在高温高压下可起化学反应,因此技术部不建议将葡萄糖加入后高压,而建议用上述提示1的方法配制。

3 建议 : 棉子糖和半乳糖原则上是不能高温高压的,因为在高温高压下其化学结构会发生变化从而对诱导效果产生抑止,这一点特别提醒研究者务必注意,尤其是在做表达调节介导的功能互补实验时至关重要!

4:制备平皿即软琼脂半固体培养基(琼脂粉按2%即每100ml加2克琼脂粉)用于转化筛选时,高压前需要将pH调至6.0左右(为方便起见,pH范围可在5.6-6.5之间变化),而液体培养基则不需要调pH。
