克隆号3H3单克隆抗体与小鼠4-1BB反应,小鼠4-1BBTNF受体超家族成员,也称为CD1374-1BB是由T淋巴细胞、NK细胞、树突细胞、粒细胞和肥大细胞表达的39 kDa跨膜蛋白。在结合其配体4-1BBL后,4-1BB通过激活NF-κBc-Junp38下游途径向CD4CD8 T细胞提供共刺激信号。4-1BB途径的重要性在包括癌症在内的许多疾病中得到了强调。据文献报道,激动性抗4-1BB抗体可诱导T细胞介导的抗肿瘤免疫。克隆号3H3抗体是一种激动性抗体,已显示在体内刺激4-1BB信号传导。

克隆号145-2C11单克隆抗体能与小鼠CD3ε反应,CD3ε是一种属于免疫球蛋白超家族的分子量为20 kDa跨膜细胞表面蛋白。CD3ε是结合形成TCR复合物的五条多肽链之一。CD3ε在T淋巴细胞、NK-T细胞和发育中的胸腺细胞上均有不同程度的表达。CD3在TCR信号传导、T淋巴细胞活化和抗原识别中起作用。克隆号145-2C11抗体已被证明通过结合和刺激TCR在体外诱导T淋巴细胞活化、增殖和凋亡。据报道,当在体内使用时,该抗体会产生T细胞活化、无反应或死亡。体内静息T细胞的激活导致细胞因子的释放和随后由T细胞和含Fcγ受体(FcR)的细胞的Ab介导的交联引起的毒性。因此,非FcR结合145-2C 11 f(ab’)2片段更常用于体内应用。据报道,克隆号145-2C11抗体可阻断克隆号17A2抗体与CD3ε+ T淋巴细胞的结合。

JC071单克隆抗体能与犬PD-L1(程序性死亡配体1,也称为B7-H1或CD274)发生反应。PD-L1是一种40kDa的I型跨膜蛋白,属于Ig超家族的B7家族。PD-L1在T淋巴细胞、B淋巴细胞、NK细胞、树突状细胞以及IFN-γ刺激的单核细胞、上皮细胞和内皮细胞上表达。PD-L1与其受体PD-1结合,PD-1存在于CD4和CD8胸腺细胞以及活化的T、B淋巴细胞和髓细胞上。PD-L1与PD-1的结合抑制TCR介导的T细胞增殖和细胞因子产生。PD-L1被认为在肿瘤免疫逃逸中起着重要作用。诱导的PD-L1表达在许多肿瘤中很常见,并导致肿瘤细胞对CD8阳性 T细胞介导的裂解的抗性增加。JC071抗体可以通过流式细胞术和蛋白质印迹检测犬组织上的PD-L1。

FGK4.5-CP133单克隆抗体能与小鼠CD40(也称为Bp50)反应。CD40是一种48 kDa的I型跨膜糖蛋白,属于肿瘤坏死因子受体(TNFR)超家族。CD40广泛表达于抗原呈递细胞(APCs),如树突状细胞、B细胞、巨噬细胞和单核细胞,以及非免疫内皮细胞、基底上皮细胞和一系列肿瘤细胞上。CD40与其配体CD154结合后,作为共刺激分子激活B细胞、树突状细胞、单核细胞和其他APCs。CD40在B细胞活化、分化、增殖和免疫球蛋白同种型转换以及树突状细胞成熟中起作用。激动性CD40单克隆抗体已被证明能激活APCs并促进抗肿瘤T细胞反应。FGK4.5单克隆抗体是一种激动性抗体,已被证明能激活表达CD40的APC。FGK4.5单克隆抗体也可用于体外和体内抑制CD40/CD154相互作用。

DC101-CP132单克隆抗体是原始DC101单克隆的重组嵌合型抗体。可变结构域序列与原始DC101相同,但是恒定区序列已经从大鼠IgG1变为小鼠IgG2a。DC101-CP132单克隆抗体像原始大鼠IgG1抗体一样,不包含Fc突变。

 

DC101-CP132单克隆抗体能与小鼠VEGFR-2(血管内皮生长因子受体2)反应,VEGFR-2也称为CD309、KDR和Flk-1。VEGFR-2是酪氨酸蛋白激酶家族的成员。当结合到其配体血管内皮生长因子时发挥生理作用,血管内皮生长因子受体-2在血管发育和通透性中起关键作用。血管内皮生长因子受体-2在成人心脏、肺、肾、脑和骨骼肌的内皮细胞中高表达,在其他组织中低表达。DC101单克隆抗体已被证明在体内抑制VEGFR-2信号传导。
