◆摘要

 

    （蛋白质工程学试剂）（氨基酸序列分析试剂）（序列分析酶）

      在温和条件下进行蛋白多肽链的切断，不会引起氨基酸残基侧链的修饰（氧化、卤化、脱酰胺等）。而且，被修饰残基（糖链、类脂体、磷酸、硫酸等的结合部位）的肽一般以完整形态被回收，而且具有良好的回收率。如果采用同样的消化条件可以实现高再现性的分段。使用底物特异性高的酶进行蛋白质的结构分析，可预测氨基酸组成的大体片段数和平均大小，并且重要的是可以避免由于部分切断（非特异性切断），导致分离的复杂化。

      本品由Drapeau系列的Staphylococcus aureus（V8菌株）分离而成，是特异性切断谷酰胺产的羧基侧链的酶。分别切断磷酸系列缓冲液中包含谷氨酸及天冬氨酸羧基侧链、氨系列缓冲液中包含谷氨酸羧基侧链的肽结合酯结合。因为其高底物特异性，本品被用于各种蛋白质序列分析时的肽分段。

 

 

◆特异性

 

      切断氨缓冲液中包含谷氨酸羧基侧链的肽结合和酯结合。切断磷酸缓冲液中包含谷氨酸及天冬氨酸羧基侧链的肽结合和酯结合。 

活性：20 units/mg以上

单位的定义：以2-Phe-Leu-Glu-4-NA为底物，在pH7.8，25℃情况下，1分钟生产1 μmol 4-硝基苯胺所需酶的量为1 unit。

反应：特异性切断谷氨酸的羧基侧链。

 

 

◆使用方法

 

仅切断谷氨酸羧基

 

      在0.01~0.1 mol/L碳酸氢铵（pH7.8）或者在0.01~0.1 mol/L醋酸铵（pH4.0）中，酵素：底物（mol/mol）=1：30~100，在30~37℃情况下，分解2~24小时。

 

 

同时切断天冬氨酸羧基

 

      在0.01~0.1 mol/L磷酸缓冲液（pH7.8）中，以同样条件分解。

 

用途：各种蛋白质的序列分析的肽分段。

pH情况：

.佳pH：在pH3.5~9.5范围有蛋白酶活性。pH4.0及pH7.8时，活性大（以血红蛋白为底物）。

抑制情况：主要抑制剂：DFP

 

 

◆使用上应注意

 

稳定性

 

      pH值为4~10时，物质稳定。冷冻干燥或冷冻，保存时间较长。在0.2%的SDS中，具有100%活性：在4 mol/L尿素中，具有50%活性。
