ChromaLink生物偶联技术是基于由芳香肼和芳香醛形成稳定的双芳基腙。S-HyNic(琥珀酰亚胺基6-肼基烟酸丙酮腙，SAH)用于在生物分子上掺入芳香肼部分。S-HyNic是一种胺反应试剂，直接将生物分子和表面的氨基转化为HyNic基团。S-4FB(琥珀酰亚胺基4-甲酰苯甲酸酯，SFB)用于将氨基转化为芳香醛(4-甲酰苯甲酰胺(4FB)基团)。向4FB修饰的生物分子或表面添加HyNic修饰的生物分子直接导致缀合物的形成(图1)。双芳基腙键对92O和pH 2.0-10.0稳定。由于抗体在低酸碱度(即< 5.0)下免疫反应的不稳定性，抗体结合的推荐酸碱度为6.0。与基于硫醇的偶联协议不同，基于硫醇的偶联协议需要还原剂，还原剂可以通过切割二硫键损害蛋白质的活性，HyNic-4FB偶联使二硫键保持完整。共轭物的制备不需要氧化剂、还原剂或金属。

CLEARLY Stained Protein Ladder是由11种高纯度预染型重组蛋白质组成的混合物。经SDS-PAGE电泳检测，可以看到从10-210 kDa清晰可辨的条带。30 kDa和70 kDa条带是其他条带质量的4倍，可以作为指示带。
本制品已经预混了Loading Buffer，可直接上样电泳，不需要预热处理。使用SDS-PAGE minigel时，以每个泳道上样10 μl，本制品可使用50次。
本制品用于监测SDS-PAGE中的蛋白质迁移和验证Western印迹*中的转移效率。
 *相比于未染色的蛋白质，由于染料和蛋白质的共价偶联会影响SDS-PAGE胶上的电泳迁移，因此，本制品不建议作为精确的分子量大小衡量标准来确定目的蛋白质的分子量。

Product Information

 The Monarch Protein Separation Solution, supplied as a component of the Monarch HMW DNA Extraction Kit for Tissue (NEB #T3060), is a salt-based solution that is optimized to ensure proteins in tissue and bacteria samples are easily removed during the phase separation of the Monarch HMW DNA Extraction workflow. It is supplied in two bottles of 18 ml.
