检验原理
蛋白胨、酵母浸粉提供基本的营养;高浓度的氯化钠具有选择性,可抑制部分不耐盐细菌的生长;甘露醇为可发酵糖类;磷酸氢二钾为pH缓冲剂;苯酚红为酸碱指示剂;琼脂为凝固剂;较高浓度的亚碲酸钾可抑制多数杂菌的生长,并可被金黄色葡萄球菌还原,菌落呈黑色。
金黄色葡萄球菌具有较强的耐盐性,可在此培养基中生长,并还原亚碲酸钾,菌落显黑色,同时可分解培养基中的甘露醇产酸,使培养基变黄。
用法:称取本品 90.0g,加热溶解于 1000ml 蒸馏水中,121℃高压灭菌 15 分钟,冷至50℃左右时,加入无菌 1%亚碲酸钾溶液 15ml,混匀,倾入无菌平皿,备用。
贮存方法:请放置阴凉干燥处保存。
金黄色葡萄球菌在TMP琼脂培养基上的生长特征:

按标签用法制备培养基,接种以下质控菌株,放置36±1℃需氧培养24-48小时。
注:回收率计算时,用TSA琼脂做对照培养基。
