In vitro(体外研究)
PD0166285 is identified to inhibit Wee1 activity at nanomolar concentrations. The inhibitor abrogates G2/M checkpoint inducing early cell division. At the cellular level, 0.5 μM PD0166285 dramatically inhibits irradiation-induced Cdc2 phosphorylation at the Tyr-15 and Thr-14 in seven of seven cancer cell lines tested. This G2 checkpoint abrogation by PD0166285 is demonstrated to kill cancer cells.PD0166285 does not inhibit Cdc2/cyclin B but inhibits Chk1 kinase at a much higher concentration (3433 nM). the treatment of cells with the inhibitor is related to microtubule stabilization and decrease in cyclin D transcription. Thus, PD0166285 may be a potentially useful anti-cancer therapy.
In vivo(体内研究)
PD0166285 at 0.5 μM concentration can inhibit Cdc2Y15 /T14 phosphorylation in all cell lines tested, regardless of their p53 status and pharmacological targeting of WEE1 by PD0166285 sensitizes U251-FM GBM tumors to IR in vivo.

概述

NEB 提供几种表达载体，该载体能表达携带有 SNAP-tag 和 CLIP-tag 的融合蛋白，适用于在哺乳动物及细菌中标记，相应的启动试剂盒也包含有相应的载体。哺乳动物 SNAPf 和 CLIPf 载体表达 SNAP- 和 CLIP-tags 的速度较之前的载体更快。表达载体中，tag 的上、下游具有改造后的多克隆位点，使得 tag 可以表达在目的蛋白的任何一端，该表达过程受 CMV 启动子调控。SNAPf-tag 和 CLIPf-tag 表达载体中携带新霉素抗性基（NeoR），可用于筛选稳定转染子。载体上还带有一个 IRES 元件，有助于融合蛋白和 NeoR 的高效表达。为在哺乳动物细胞中高效表达，Tag 基因中的密码子已经过优化。NEB 还提供对照质粒，这些质粒编码的融合蛋白分别定位于细胞（H2B）、线粒体（Cox8A）和细胞表面（ADRβ2, NK1R），启动试剂盒中包含相应的对照质粒（见 290 页）。
 
细菌表达载体 pSNAP-tag(T7)-2 包含位于 SNAP-tag 上、下游的克隆位点，受 T7启动子调控。为在E. coli 中高效表达，SNAP-tag 基因中的密码子已经过优化。

通过标准的质粒纯化程序，分离自 E. coli 菌株。质粒无内毒素污染，可用于高效转染。

SMARTer Stranded Total RNA Sample Prep Kit – Low Input Mammalian (Code No. 634861) -从总RNA到RNA-seq文库的完整解决方案，适用于大部分人、大鼠或小鼠样品。
SMARTer Stranded RNA-Seq Kit HT(Code No. 634862)-采用Illumina高通量index构建96种index RNA-seq文库。其他SMARTer Stranded RNA-Seq Kit采用12种低通量index。
SMARTer Stranded RNA-Seq Kit 提供了一种构建Illumina链特异性测序文库的解决方案，并且链特异性识别准确率>99%，可以从低至100 pg至100 ng的RNA起始获得可靠数据。SMARTer Stranded RNA-Seq Kit 采用随机引物法适用于原核或真核RNA，推荐使用去除rRNA或poly（A）富集RNA样品。本试剂采用改良的核心SMART技术，可以生成完整转录组文库，包括编码和非编码RNA，同时添加index和接头，无需下游文库构建试剂。采用SMARTer Stranded RNA-Seq Kit不到4小时即可构建适用于Illumina测序的index文库。同时提供SMARTer Stranded RNA-Seq Kit HT，包括8条正向引物和12条反向引物可以生成多至96个独立index文库用于高通量研究。
对于10-100 ng总RNA中的rRNA去除，推荐使用RiboGone – Mammalian kit。 SMARTer Stranded Total RNA Sample Prep Kit – Low Input Mammalian 结合了SMARTer Stranded技术和RiboGone rRNA去除技术，可以实现从总RNA样品起始到RNA-seq文库制备的无缝衔接，5 h内即可完成。
