CUTANA pAG-Tn5是靶向剪切及转座酶(CUT&Tag)技术中进行高效绘制染色质特征的关键试剂。与ChIP-seq相比,CUT&Tag在降低细胞需求量和测序深度的信噪比方面进行了显著改进。CUTANA pAG-Tn5是一种高活性的E. coli转座酶突变体(Tn5)与蛋白A/G的融合产物,可与来自多种物种宿主的靶抗体兼容。它也没有表位标签,使其适合于标签介导的CUT&Tag(如FLAG、HA、TY1、V5等)。该产品经过高度纯化,已去除污染的E. coli DNA,并在细胞数量少的情况下进行复杂分析。为了每次实验的标准化以及抗体验证和反应监测,推荐使用SNAP-CUTANA™ nucleosome spike-ins(如EpiCypher 19-1002)。Tn5带有mosaic 接头,可在CUT&Tag中使用。

产品简介:

SDS-PAGE蛋白上样缓冲液((SDS-PAGE Sample Loading Buffer, 6×),是一种经过改良的以溴酚蓝为染料的6倍浓缩的蛋白上样缓冲液,可以用于常规的SDS-PAGE蛋白样品的上样。

本缓冲液已经含有还原剂DTT,有轻微刺激性气味。

 

使用方法:

1、在常温或不超过37℃的水浴中溶解SDS-PAGE上样缓冲液(6x)。水浴溶解后立即常温存放,尽量避免长时间置于水浴中。

2、按照每5ul蛋白样品加入1ul蛋白上样缓冲液(6×)的比例,混合蛋白样品和蛋白上样缓冲液(6×)

3、95℃~100℃或沸水浴加热10分钟,以充分变性蛋白。

4、冷却到常温后,直接上样到SDS-PAGE凝胶胶加样孔内即可。

5、通常电泳至蓝色染料到达胶的底端处附近即可停止电泳。

 

注意事项:

1、样品加热前通常会发现蛋白样品内有粘稠的半透明状物体,通常在上样缓冲液内加热8-10分钟后消失。如果起始时细胞或组织的用量较大,基因组DNA含量较高,加热10分钟后有可能仍然比较粘稠或者有粘稠状的半透明物体。此时需要再煮沸10分钟或者补加适量6x的蛋白上样缓冲液后再煮沸5分钟。充分煮沸后一方面可以使结合在基因组DNA上的蛋白充分释放,同时会导致基因组DNA的部分断裂从而使粘稠感消失,这样就不会影响后续的上样操作。

2、SDS-PAGE蛋白上样缓冲液(6x)必须完全溶解后再使用,应避免反复冻融。

3、本产品仅限于专业人员的科学研究用,不得用于临床诊断或治疗,不得用于食品或药品。
