产品用途

酵母是常用的模式生物,经常用来进行基因改造,而酵母氨基酸缺陷培养基是酵母转化和酵母杂交等所必须用到的培养基,其主要用途:用于酵母单杂交/酵母双杂交等的实验研究以及酵母遗传突变株的筛选等研究。根据缺陷氨基酸的种类,分为四大类培养基,分别为一缺、二缺、三缺、四缺培养基,本公司可以根据您的要求定制相关培养基,优质保证。

二缺培养基:此类酵母SC选择培养基-SD培养基用于双质粒酵母转化筛选。

用途:外源基因在酵母中的表达、异源基因功能互补、酵母单杂交系统和酵母双杂交系统初始质粒转化。

免疫共沉淀(Coimmunoprecipitation)、Y187和AH109 酵母交配实验、接合型二倍体筛选。

注意事项

1: 上述培养基含有各种所需成分(葡萄糖除外,因为有些研究者需要用其他糖类如用半乳糖诱导GAL1启动子表达),为各种氨基酸和酵母含氮碱/酵母氮源的精细混合物干粉,按照8g/L称取,加水后即可配制成相应的培养液,无需再购置添加Minimal SD Base或其他酵母氮源成分。高压灭菌后加无菌葡萄糖至终浓度为2%(或棉子糖)或实验所需的特定诱导物(如半乳糖用于GAL1启动子诱导表达的剂量或浓度为2%)即可,适用于外源基因在酵母中的表达,酵母单杂交(yeast one-Hybrid)和酵母双杂交(yeast two-Hybrid)系统等的选择缺陷型培养,筛选含特定标志基因的酵母表型。

2: 在配制酵母缺陷培养基时,可以先加入葡萄糖后再高压,对酵母的生长不会产生严重影响。但如加入葡萄糖后再高压消毒则培养基会变色,由浅黄到深褐色不等,对细胞的生长还是有一定影响的(但在一般情况下不会影响实验)。建议: 由于氨基酸和葡萄糖在高温高压下可起化学反应,因此技术部不建议将葡萄糖加入后高压,而建议用上述提示1的方法配制。

3 建议 : 棉子糖和半乳糖原则上是不能高温高压的,因为在高温高压下其化学结构会发生变化从而对诱导效果产生抑止,这一点特别提醒研究者务必注意,尤其是在做表达调节介导的功能互补实验时至关重要!

4:制备平皿即软琼脂半固体培养基(琼脂粉按2%即每100ml加2克琼脂粉)用于转化筛选时,高压前需要将pH调至6.0左右(为方便起见,pH范围可在5.6-6.5之间变化),而液体培养基则不需要调pH。

溶解方法
Always centrifuge tubes before opening. Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100 μg/ml. Dissolve the lyophilized protein in ddH2O. Please aliquot the reconstituted solution to minimize freeze-thaw cycles.

Lyophilized protein should be stored at < -20°C, though stable at room temperature for 3 weeks. Reconstituted protein solution can be stored at 4-7°C for 2-7 days. Aliquots of reconstituted samples are stable at < -20°C for 3 months.

Human β-Nerve Growth Factor (β-NGF) was initially isolated in the mouse submandibular gland. It is composed of three non-covalently linked subunits α, β, and γ; it exhibits all the biological activities ascribed to NGF. It is structurally related to BDNF, NT-3 and NT-4 and belongs to the cysteine-knot family of growth factors that assume stable dimeric structures. Β-NGF is a neurotrophic factor that signals through its receptor β-NGF, and plays a crucial role in the development and preservation of the sensory and sympathetic nervous systems. Β-NGF also acts as a growth and differentiation factor for B lymphocytes and enhances B-cell survival. These results suggest that β-NGF is a pleiotropic cytokine, which in addition to its neurotropic activities may have an important role in the regulation of the immune system. Human β-NGF shares 90% sequence similarity with mouse protein and shows cross-species reactivity.
