醛固酮(Aldosterone)是一种由肾上腺皮质中的胆固醇合成的类固醇激素。醛固酮在肾脏和肝脏中代谢,并作为控制钠钾平衡的关键盐皮质激素发挥作用。肾上腺合成和释放醛固酮主要受肾素-血管紧张素-醛固酮系统(RAAS)的调节,RAAS主要作用是调节和维持血压、水电解质平衡和维持人体内环境稳态。检测血清醛固酮和血浆肾素在临床上用于区分原发性和继发性醛固酮增多症。原发性醛固酮增多症(醛固酮增多症)的特征是肾素与醛固酮比率非常低,导致钠滞留和血压升高,通常是肾腺增生或肿瘤的结果。在继发性醛固酮增多症中,醛固酮的过量产生是由心力衰竭和肾动脉疾病等外部条件引起的。

生物活性 Aldosterone 是原发性盐皮质激素。Aldosterone 是一种类固醇激素,通过肾上腺皮质的肾小球区 (ZG) 对肾素-血管紧张素系统激活 (RAS) 或高钾饮食反应而合成和分泌。Aldosterone 活性在细胞水平上依赖于胞浆/核盐皮质激素受体 (MR) 的结合和激活。[1-5]
In Vitro 醛固酮(1-1000nm;24小时)以剂量依赖的方式抑制白细胞介素-1β刺激的血管平滑肌细胞产生亚硝酸盐[3]。
In Vivo 醛固酮(1 mg/Kg+1%NaCl;i.h.;每日1次,持续3周)显著升高收缩压(SBP)、舒张压(DBP)、左室收缩压(LVSP)和左室舒张末压(LVEDP)[4]。醛固酮(0.72 mg/kg/天;14天)使雄性小鼠血压略微升高(14 mmHg)[5]。动物模型:40只雄性Wistar大鼠[4]剂量:1 mg/Kg(+1%NaCl)给药:i.h;每日1次,连续3周。结果:醛固酮盐组大鼠收缩压(SBP)、舒张压(DBP)、左室收缩压(LVSP)和左室舒张末压(LVEDP)明显升高。
