支原体污染是细胞培养中一个主要的污染问题,能够严重影响实验结果的重现性和有效性。InvivoGen公司凭借40多年为科学界开发支原体解决方案的经验,开发了简单可靠的检测细胞培养支原体污染的MycoStrip™试剂盒,基于等温PCR的技术检测支原体污染,在5分钟内就能于免疫层析试纸条上快速显示结果。

支原体属于柔膜菌纲,其成员特征为缺乏细胞壁以及拥有多变的形态。支原体对包括原代细胞在内的所有真核细胞类型具有高度传染性。成百上千的支原体可以附着在一个细胞上,与细胞膜融合、繁殖,最终数量可以超过培养细胞1000倍。支原体没有细胞壁的特点使它们对常用的抗生素(如青霉素和链霉素)产生抗性。 此外,它们的尺寸微小(~100 nm),能够穿透过 0.2 μm 的标准过滤膜。支原体与宿主细胞竞争营养素和代谢物前体,它们能改变许多细胞功能,影响到细胞代谢和细胞生长,最终导致细胞死亡。因此,我们需要采取预防措施来防止细胞培养物受到污染。


目前的支原体检测方法大多费时费力。Japanese cell bank验证了由InvivoGen开发的基于等温PCR的支原体快速检测试剂盒MycoStrip的有效性,并将其检测灵敏度与其他检测方法进行了比较。实验中,将M. hyorhinis支原体(NBRC 14858)加入到VERO细胞(JCRB0111)中进行培养,稀释后的样品分别用MycoStrip、生物发光法(Bioluminescence assay)、荧光染色法(Fluorescent staining)和Nested-PCR进行检测。经证实,MycoStrip的灵敏度与荧光染色的灵敏度相当,显示出足够的检测效率证明了基于等温PCR的MycoStrip是一种简单且高度通用的支原体检测方法,因为该检测方法不需要特殊设备,将试剂与样品混合并滴在试纸上即可轻松快速地获得结果。MycoStrip可普遍用于支原体污染的常规检测。

常见问题

1. 如果支原体检测呈阳性怎么办?

InvivoGen有解决方案!可以使用InvivoGen全面的抗支原体试剂清除和预防污染。推荐用Plasmocin™ 或Plasmocure™ ,治疗完成后(约2周),使用MycoStrip™重新检测。 

2. 收集的待测上清液中含有一些细胞,会干扰检测吗?

无需担心,哺乳动物细胞DNA的存在不会影响检测分析。 

3. 样品制备过程中,将样品加热到95°C是否有好处?

敏感度没有提高,但此步骤可以杀死支原体,避免接触活的支原体。

4. 是否每次使用MycoStrip™时都需要设立阳性和阴性对照?

包括阳性和阴性对照的定期监测,对于结果的可靠性以及故障排除很重要。

5.在什么阶段可以冻存样品以供后续检测分析?

可以按照以下方式储存样本:

• 细胞培养上清液:在开始“样品制备”方案之前,可以将上清液储存在-20°C直到需要使用时。使用前,将样品在37°C下完全解冻,涡旋充分混匀以确保所有盐溶解。

• 已制备的样品:执行“样品制备”方案的步骤1-4之后,可以将样品存储在-20°C直至需要使用时。 

• 处理过的样品:执行“检测分析”方案的步骤1-4之后,处理过的样品在-20°C下最多保存3个月。

6. 有一个样本的测试带模糊,不能确定是否应将其视为支原体污染的阳性结果?

如果其中一个样品上观察到一条微弱的条带,可以认为这是支原体污染的阳性结果。建议使用以下方法确认此结果: 

• 浓缩样品:使用更大体积的上清液并重复“样品制备”方案中步骤2-3。使用MycoStrip™重新检测浓缩后的样品。

• 在使用MycoStrip™重新检测之前,继续培养细胞48小时。

• 使用无菌水(不含核酸酶)替代PBS重悬样品,以提高检测的灵敏度。

7. 实验室中没有热循环仪,可以使用什么代替?

可以使用校准过的加热块或水浴代替。请注意,使用水浴可能会引入新的污染源。 

8. 如果将准备好的样品在加热块上放置了一个多小时,是否仍然能使用该样品?

在65°C下孵育40分钟以上会显著影响检测结果,因此不建议继续使用此样品。

腺苷(Adenosine)是一种嘌呤核苷,包含通过β-N9糖苷键连接到核糖分子(呋喃核糖)的腺嘌呤部分。腺苷衍生物在能量转移反应(如三磷酸腺苷(ATP)和二磷酸腺苷(ADP))以及信号转导(如环磷酸腺苷(cAMP))中发挥重要作用。此外,腺苷是一种神经调节剂,被认为可以促进睡眠和抑制觉醒。腺苷还通过血管舒张调节流向多个器官的血流。腺苷是腺苷高胱氨酸酶(AHCY)将S-腺苷高半胱氨酸(SAH)转化为高半胱氨酸的副产物。

Cell Biolabs的腺苷检测试剂盒是一种简单的荧光测定法,可在96孔板中测量生物样品中存在的总腺苷量。检测原理是腺苷通过腺苷脱氨酶(ADA)转化为肌苷。然后肌苷被嘌呤核苷磷酸化酶(PNP)转化为次黄嘌呤。最后,次黄嘌呤被黄嘌呤氧化酶(XO)转化为黄嘌呤和过氧化氢。然后用高特异性荧光探针检测生成的过氧化氢。辣根过氧化物酶催化探针和过氧化氢之间的反应,它们以1:1的比例结合。将样品与96孔板中已知浓度的腺苷标准品进行比较。将样品和标准品孵育15分钟,然后用标准的96孔荧光酶标仪测定读数。该试剂盒的检测灵敏度为1.56 uM

500,000 K562 cells per sample were processed with the CUTANA™ ChIC/CUT&RUN Kit (EpiCypher 14-1048) following treatment with IgG negative control (EpiCypher 13-0042), H3K4me3 (EpiCypher 13-0041), H3K27me3 (EpiCypher 13-0030) and CTCF (EpiCypher 13-2014) antibodies. Isolated DNA was used to prepare paired-end Illumina® sequencing libraries, which were analyzed by Agilent TapeStation® , confirming enrichment of mononucleosomes (~300 bp peak represents ~170 bp excised DNA + sequencing adapters).

Cleavage Under Targets & Release Using Nuclease (CUT&RUN) utilizes an immunotethering approach to specifically release genomic fragments of interest into solution, enabling next-generation sequencing and genomic mapping with unprecedented sensitivity. High yield purification and concentration of partitioned chromatin DNA is an essential step – one that requires a workflow specifically compatible with low starting material. Utilizing tapered cleanup columns designed for low elution volumes (as low as 6 µL), this kit produces purified, concentrated DNA ready for library preparation and next-generation sequencing (Figure 1). The user-friendly workflow can be completed in just 30 minutes. The columns recover > 50 bp DNA fragments, making the DNA Purification Kit ideal for a variety of CUT&RUN targets, including histone PTMs and transcription factors.
