TLR3(又名CD283)是一个胞内受体,可识别与病毒感染相关的双链RNA(dsRNA)分子模式。其中一个TLR3的配体,Poly(I:C),是人工合成的dsRNA类似物。TLR3信号通路不依赖于MyD88,而是通过接头蛋白TRIF(包含TIR结构域的接头蛋白,可诱导IFN-β产生)发挥作用。IRF3(干扰素调节因子3)被磷酸化后导致IFN-β(β干扰素)产生,进一步活化ISRE(干扰素激活反应元件)。另外,TLR3信号通路的激活也可导致AP-1和NF-kB等转录因子入核,促进一些炎症因子的释放。

克隆号FGK4.5单克隆抗体能与小鼠CD40(也称为Bp50)反应。CD40是一种48 kDa的I型跨膜糖蛋白,属于肿瘤坏死因子受体(TNFR)超家族。CD40广泛表达于抗原呈递细胞(APCs),如树突状细胞、B细胞、巨噬细胞和单核细胞,以及非免疫内皮细胞、基底上皮细胞和一系列肿瘤。CD40与其配体CD154结合后,作为共刺激分子激活B细胞、树突状细胞、单核细胞和其他APCs。CD40在B细胞活化、分化、增殖和免疫球蛋白同种型转换以及树突状细胞成熟中起作用。激动性CD40单克隆抗体已被证明能激活APCs并促进抗肿瘤T细胞反应。克隆号FGK4.5抗体是一种激动性抗体,已被证明能激活表达CD40的APC。克隆号FGK4.5也可用于体外和体内抑制CD40/CD154相互作用。

克隆号145-2C11单克隆抗体能与小鼠CD3ε反应,CD3ε是一种属于免疫球蛋白超家族的分子量为20 kDa跨膜细胞表面蛋白。CD3ε是结合形成TCR复合物的五条多肽链之一。CD3ε在T淋巴细胞、NK-T细胞和发育中的胸腺细胞上均有不同程度的表达。CD3在TCR信号传导、T淋巴细胞活化和抗原识别中起作用。克隆号145-2C11抗体已被证明通过结合和刺激TCR在体外诱导T淋巴细胞活化、增殖和凋亡。据报道,当在体内使用时,该抗体会产生T细胞活化、无反应或死亡。体内静息T细胞的激活导致细胞因子的释放和随后由T细胞和含Fcγ受体(FcR)的细胞的Ab介导的交联引起的毒性。因此,非FcR结合145-2C 11 f(ab’)2片段更常用于体内应用。据报道,克隆号145-2C11抗体可阻断克隆号17A2抗体与CD3ε+ T淋巴细胞的结合。
